Sibelium is used for managing different kind of migraine (i.e. with aura and without aura) and certain type of vertigo problems. The active ingredient present in Sibelium is flunarizine which belongs to the chemically heterogeneous group of calcium-entry blockers. The mechanism of relieving pain is based on prevention of cellular calcium surplus by reducing unnecessary trans membrane calcium influxes. It is sparingly soluble in water and it reaches peak plasma levels within 2 – 4 hours of intake and attains steady state in period of 5 to 6 weeks. Flunarizine is bound for 99.1% to human plasma proteins, and the blood/plasma concentration ratio is 0.60. It is excreted through the faeces after general hepatic metabolism. The mean terminal elimination half-life of flunarizine is of approximately 18 days.
